Subsequently, the researchers used different types of DDR inhibitors to further study the relationship between DNA damage repair mechanism and SARS-CoV-2 virus infection. The results showed that the three DDR inhibitors inhibited the virus infection to varying degrees. It shows that both DNA damage and DDR repair mechanisms can promote the infection of the new coronavirus.

Based on the experimental data, the researchers speculated that both the new coronavirus and the SARS virus use the ACE2 receptor on the cell surface to enter the cell, and verified it. found that ACE2 expression increased after ionizing radiation-induced DNA damage. In contrast, ionizing radiation-induced ACE2 expression was reduced after DDR inhibitors were administered. It shows that the cause of virus infection is very likely to be closely related to ACE2. The results are obviously consistent with the conjecture.

On this basis, researchers fed 12-month-old mice with 200 mg/kg/day of NMN for 7 days. The mice were also induced to be infected with the new coronavirus, and the expression of γH2AX, ACE2 and p-c-Jun was found to be reduced, and the viral load and tissue damage in the lungs were also significantly reduced. Shows that NMN really works!

This study explains why the elderly are susceptible to the new crown from the perspective of DNA damage. The fly in the ointment is that this study is still limited to mice. If it can be verified clinically in humans, it will be of great significance.
