On January 25, 2023, a research team from Kobe Institute of Biomedical Research, Tokyo Institute of Aging, and Washington University School of Medicine published a research report in the international research journal "NPJ Aging", proving that aging animals restore the arcuate nucleus of the hypothalamus by taking NMN (Neurons gathered at the base of the medial thalamus), ventromedial hypothalamus (injury causes obesity and gonadal atrophy), lateral hypothalamus (responsible for controlling appetite and regulating satiety) NAD+ levels to young levels, further proving that taking NMN is effective anti-aging method.
The hypothalamus has been shown to be a higher-order control center of aging in mammals, and two subregions within the hypothalamus, the dorsal and lateral hypothalamic nuclei, play key roles in regulating aging and longevity in mammals. In these two subregions, NAD+-dependent protein deacetylase Sirt1 and homeodomain transcription factor Nkx2-1 cooperate to regulate the expression of key genes and promote neuronal activity to delay aging and extend the lifespan of mice.

The study compared the local NAD+ changes in the hypothalamic nucleus of 3-month-old mice (equivalent to 20 years in humans) and 22-month-old mice (equivalent to 60 years in humans), and found that the hypothalamic arcuate nucleus (ARC) , NAD+ levels in the ventromedial hypothalamus (VMH) and lateral hypothalamus (LH), significantly decreased in 22-month-old mice.

After the researchers gave 300mg/kg of NMN to aged mice, they found that the arcuate nucleus of the hypothalamus (ARC, neurons gathered at the base of the medial thalamus), the ventromedial hypothalamus (VMH, injury-induced obesity and gonadal atrophy), the lateral hypothalamus (LH, responsible for controlling appetite and regulating satiety) and NAD+ levels in the dorsomedial hypothalamic nucleus (DMH, stress-induced cardiovascular response) were significantly increased.

The results of this study suggest that DNA damage and inflammation lead to a decline in thalamus function and NAD+ levels with age, which in turn induces insomnia, physical activity and cognitive decline. By taking NMN to supplement NAD+, it is possible to restore and maintain the NAD+ level when you were young, and significantly delay the symptoms of aging, thereby improving sleep, mobility, and cognitive abilities.
